Ventrolateral periaqueductal gray GABAergic neurons promote arousal of sevoflurane anesthesia through cortico-midbrain circuit

چکیده

The mechanism of general anesthesia remains elusive. ventrolateral periaqueductal gray (vlPAG) in the midbrain regulates sleep and awake states. However, role vlPAG its circuits is unclear. We utilized opto/chemogenetics, righting reflex, electroencephalographic recording to assess consciousness changes. employed fiber photometry measure activity neurons neurotransmitters. As a result, showed that GABA decreased during sevoflurane was reactivated after anesthesia. Activating GABAergic promoted arousal anesthesia, while inhibiting them delayed this process. Furthermore, medial prefrontal cortex (mPFC) pyramidal projections ventral tegmental area (VTA) played prominent anesthesia-awake transition. neurotransmitter VTA synchronized with mPFC-vlPAG neuron projections. Therefore, cortico-midbrain centered on exert an arousal-promoting effect

Stimulation of the Midbrain Periaqueductal Gray Modulates Preinspiratory Neurons in the Ventrolateral Medulla in the Rat In Vivo

The midbrain periaqueductal gray (PAG) is involved in many basic survival behaviors that affect respiration. We hypothesized that the PAG promotes these behaviors by changing the firing of preinspiratory (pre-I) neurons in the pre-Bötzinger complex, a cell group thought to be important in generating respiratory rhythm. We tested this hypothesis by recording single unit activity of pre-Bötzinger...

The midbrain periaqueductal gray control of respiration.

The midbrain periaqueductal gray (PAG) organizes basic survival behavior, which includes respiration. How the PAG controls respiration is not known. We studied the PAG control of respiration by injecting D,L-homocysteic acid in the PAG in unanesthetized precollicularly decerebrated cats. Injections in different parts of the PAG caused different respiratory effects. Stimulation in the dorsomedia...

PERIAQUEDUCTAL GRAY 1 Running head: PERIAQUEDUCTAL GRAY Common representation of pain and negative emotion in the midbrain periaqueductal gray

Human neuroimaging offers a powerful way to connect animal and human research on emotion, with profound implications for psychological science. However, the gulf between animal and human studies remains a formidable obstacle: Human studies typically focus on the cortex and a few subcortical regions such as the amygdala, whereas deeper structures such as the brainstem periaqueductal gray (PAG) p...

Neurochemical properties of BDNF-containing neurons projecting to rostral ventromedial medulla in the ventrolateral periaqueductal gray

The periaqueductal gray (PAG) modulates nociception via a descending pathway that relays in the rostral ventromedial medulla (RVM) and terminates in the spinal cord. Previous behavioral pharmacology and electrophysiological evidence suggests that brain-derived neurotrophic factor (BDNF) plays an important role in descending pain modulation, likely through the PAG-RVM pathway. However, detailed ...

Ebf2 is required for development of dopamine neurons in the midbrain periaqueductal gray matter of mouse.

Dopaminergic (DA) neurons in the midbrain ventral periaqueductal gray matter (PAG) play critical roles in various physiological and pathophysiological processes including sleep-wake rhyme, antinociception, and drug addiction. However, the molecular mechanisms underlying their development are poorly understood. Here, we showed that PAG DA neurons arose as early as E15.5 in mouse embryos. During ...
